Abstract
Ainitial investigation using gas chromatography-mass spectrometry (GC-MS) to investigate the nicotine comprised in tobacco departs was carried out. Nicotine in commercially available tobacco and tobacco departs was extracted with methanol and very resolute by GC-MS. The detection limit was at the PPM level for nonselective monitoring and the nano-gram level for nicotine selective detection. This scheme supplied a easy analytical method for the analysis of nicotine in tobacco leave. Compared to currently utilized procedures, the GC-MS provided benefits of high sensitivity, nicotine specific detection and smaller instrumentation cost.

Chapter 1: Introduction
Background of the study
A study finished by the Centre for Health Policies and Services in United Kingdom displayed that 39.9 per hundred of London smokes daily. An initial investigation using gas chromatography-mass spectrometry (GC- MS) to investigate the nicotine contained in tobacco leaves will be conveyed out. Nicotine in commercially accessible cigarettes and tobacco leaves was extracted with methanol and very resolute by GC-MS. (Bonner 2007 26)
Problem Statement
The difficulty declaration for this study would be to find the best way to evolve for determination of nicotine from tobacco leaves.
Research Aims and Objectives
? to find the best accessible way to extract nicotine from the tobacco leaves
? Whether HPLC or GC-MS procedures are better for the determination of nicotine from pharmaceutical formulations
Significance of the Study
Nicotine administered by tobacco smoking has a somewhat short length of action. Peak blood nicotine levels are accomplished normally inside the time it takes to spend the tobacco (5 to 10 minutes) and down turn quickly. Alcohol, although, by the oral path, has a slower onset and longer length of action. In this paper, a straightforward, cheap chromatographic procedure will be utilised to recount the modes for determination of nicotine in the depart and arises of tobacco.
Rationale/ Nature of the study
It is one of the highly toxic chemicals belonging to the tobacco alkaloids. Several gas chromatographic methods have been recounted for the determination of nicotine. The most routinely utilised techniques employed engage solvent extraction followed by gas chromatographic-mass spectrometric investigation or fluid chromatography (LC) with ultra-violet absorbance detection. Kim (Kim 2006 204) recounted a method for the determination of nicotine and extends in plasma after a lone extraction procedure by gas ...
